The truck left the road and ended up in a ditch the Ballarat to Maryborough railway line uses to pass under the highway.
The driver of the truck, who is yet to be formally identified, died at the scene.
Police are investigating the cause of the crash and will prepare a report for the Coroner.
The Midland Highway has been closed in both directions and buses are replacing V-Line trains between Maryborough and Ballarat.
